Overview

Humanin was the first mitochondrial-derived peptide to be described, transcribed from a short open reading frame in the mitochondrial 16S rRNA region. Research has characterized its cytoprotective activity, including suppression of apoptosis and modulation of stress-response signaling in neuronal, cardiac, and metabolic models.

Studies have examined its release by astrocytes, its relationship to lifespan and healthspan endpoints, and its behaviour within the wider mitokine literature alongside MOTS-c.

Humanin was identified in screening work seeking factors that protect neurons from amyloid-beta toxicity, and was subsequently traced to a mitochondrial open reading frame.
